贲门癌中长链非编码RNA和微小RNA差异表达谱及相关性分析
Differential expression profiles and correlation analysis of lncRNA and miRNA in gastric cardia adenocarcinoma

Abstract
Aim:To analyze the expression profiles and correlations of long non-coding RNA(lncRNA) and microRNA(miRNA) in gastric cardia adenocarcinoma (GCA) and surrounding tissue.Methods:Using microarray technology,the expression of lncRNA and miRNA in 15 pairs of GCA and matched surrounding mucosa tissue samples were profiled.The correlations between the expressions of lncRNA and miRNA were evaluated by bioinformatics approach,followed byqRT-PCR to verify the correlations.Results:A total of lncRNAs(> 5-fold difference) and 152 miRNAs (> 2-fold difference) were identified in 15 GCA samples compared with their counterparts.Among these 152 differential miRNAs,7 miRNA candidates were predicted to be targets of lncRNA H19.In another independent cohort analysis of 37 GCA cases,there was negative correlation between expression of H19 and miR-148a-3p validated by qRT-PCR (r =-0.398,P < 0.001).Conclusion:Differentially expressed lncRNAs and miRNAs are implicated in carcinogenesis of gastric cardia.LncRNA H19 may directly modulate the expression of miR-148a-3p.关键词
