Abstract

Objective To investigate the effects of miR-182-5p inhibitor combined with 5-fluorouracil (5-FU) on the proliferation,apoptosis,and migration of 5-FU-resistant human gastric cancer cell line (SGC7901/5-FU) and the sensitizing effect of miR-182-5p inhibitor on gastric cancer cells in 5-FU chemotherapy.Methods SGC7901/5-FU cells were divided into miR-182-5p oligonucleotide negative control group (group A),miR-182-5p inhibitor group (group B),5-FU group (group C),and miR-182-5p inhibitor combined with 5-FU group (group D).The proliferation,apoptosis,and migration of SGC7901/5-FU cells in each group were measured by MTT assay,flow cytometry,and Transwell assay,respectively.Results There were significant differences in survival rate,apoptosis rate,and migration rate of SGC7901/5-FU cells between group D and groups A,B,and C (F =107.38-490.53,q=4.23-48.60,P<0.05).Conclusion A combination of miR-182-5p inhibitor and 5-FU significantly reduces the proliferation and migration of SGC7901/5-FU cells,promotes cell apoptosis,and increases the sensitivity of SGC7901/5-FU cells to 5-FU chemotherapy.
