Abstract
Objective To investigate the expression of inflammatory factors tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) associated with the Toll-like receptor 4 (TLR4) signaling pathway in colorectal cancer and related clinical significance.Methods A total of 56 colorectal cancer samples and 36 colon adenoma samples were collected,and the adjacent normal intestinal mucosa samples were collected as control.Immunohistochemistry and RT-PCR were used to measure the protein and mRNA expression of TLR4,TNF-α,and IL-6,and the association of the expression of TLR4,TNF-α,and IL-6 with clinicopathological features of colorectal cancer was also analyzed.Results Colorectal cancer samples had significantly higher protein expression of TLR4,TNF-α,and IL-6 than adjacent normal tissue samples (x2 =5.250-14.756,P <0.05),as well as significantly higher mRNA expression of TLR4,TNF-α,and IL-6 than adjacent normal tissue samples and colon adenoma samples (Z =4.333-8.971,P <0.05).In colorectal cancer samples,the mRNA expression of TLR 4 was correlated with lymph node metastasis,degree of tumor differentiation,and tumor stage (r=0.304-0.338,P<0.05),and the mRNA expression of TNF-α and IL-6 was correlated with vascular invasion,lymph node metastasis,and tumor stage (r=0.269-0.320,P<0.05).Conclusion High expression of chronic inflammatory factors mediated by the TLR4 signaling pathway is closely associated with the clinicopathological features of colorectal cancer,and the TLR4 signaling pathway may play an important role in the development and progression of colorectal cancer.关键词
