Abstract
Objective To investigate the preparation of pollock skin collagen peptide-hyaluronic acid hydrogel and its influence on biological characteristics of human umbilical cord mesenchymal stem cells cultured in vitro.Methods Methacrylic anhydride,as well as fish collagen peptide and hyaluronic acid,was used to synthesize photocrosslinkable material,and human umbilical cord mesenchymal stem cells were embedded in the hydrogel for three-dimensional culture.Fluorescein diacetate staining was used to observe cell viability in the hydrogel,EdU staining and MTT assay were used to measure cell proliferation,and immunofluorescent staining was used to observe cell morphology.Results The human umbilical cord mesenchymal stem cells survived and grew well in the gel.Compared with those with adherent growth,the cells in three-dimensional culture had a spherical shape,a steady growth rate,and persistent proliferation.Conclusion Fish collagen peptide-hyaluronic acid hydrogel is suitable for cell embedding and has the potential for wide application in various fields of tissue engineering.关键词
