Screening plasma marker metabolites and the related metabolic pathways in acute myocardial infarction

Abstract
Objective To analyze the plasma metabolic profiles of patients with acute myocardial infarction(AMI)af-ter percutaneous coronary intervention(PCI), and to screen out the plasma marker metabolites and the related metabolic pathways.Methods The plasma samples from 25 patients with AMI(case group)before and after operation and 26 healthy volunteers(control group)were collected and analyzed by ultraperformance liquid chromatography-mass spectrome-try(UPLC-MS).MZmine2.0 system was used to identify,match,and normalize the ion chromatography peaks; the data were further imported into SIMCA-P+12.0.1.0 system,and the principal component analysis(PCA)and orthogonal par-tial least squares discriminant analysis(OPLS-DA)model were constructed.According to the contribution degree and con-fidence interval of the ions to the model,the potential marker metabolites were screened and the levels of the metabolites of the two groups were compared to identify the differential metabolites.The identified metabolites were imported into the MetaboAnalyst3.0 database for metabolic pathway analysis.Results The PCA model and OLPS-DA model of the two groups of plasma metabolic profiles were successfully constructed.Eight metabolites with significant differences in the plas-ma of the two groups were screened from the model.8 metabolites with significant differences in the plasma of the two groups were screened out from the models, namely, lysophosphatidylcholine(LysoPC)[18:2(9Z,12Z)], LysoPC (16:0),LysoPC(P-16:0),LysoPC(18:0);lysophosphatidic acid(LPA)[0:0/18:1(9Z)],Pregnanetriol,N-Palmi-toylsphingosine,and Cer(d18:0/16:0).Among them, the plasma levels of LysoPC[18:2(9Z, 12Z)], LysoPC (16:0),Pregnanetriol,and LysoPC(P-16:0)tended to be normal after operation,indicating that AMI and PCI treatment were important factors affecting the changes of these metabolites.The 8 differential metabolites were imported into the Meta-boAnalyst 3.0 database and the metabolic pathways associated with AMI were found, namely glycerophospholipid metabo-lism and glycerolipid metabolism.In the two metabolic pathways,there was a common metabolite which affected by AMI,a phosphatidic acid,1,2-diacyl-sn-glycerol-3-phosphate.Conclusions Eight AMI marker metabolites are screened out,in-cluding LysoPC[18:2(9Z,12Z)], LPA(0:0/18:1(9Z)), LysoPC(16:0), Pregnanetriol, LysoPC(P-16:0), N-Palmitoylsphingosine,Cer(d18:0/16:0), and LysoPC(18:0).The two metabolic pathways associated with AMI are glycerophospholipid metabolism and glycerolipid metabolism,and there is a common metabolite which is affected by AMI in both metabolic pathways,that is 1,2-diacyl-sn-glycerol-3-phosphate.关键词
