Abstract

Phosphate buffer saline(PBS)method,immunomagnetic beads adsorption method,and GB 29694—2013 method were used to extract sulfadimidine in chicken breast meat.The recovery and stability of the three methods were compared.The buffer of coupling 80 μg anti-sulfadimidine monoclonal antibody to 1 mg magnetic beads was 0.01 mol/L PBS(pH 5.0).When the dosage of immunomagnetic beads was 350 μg and the time to capture sulfadimidine was 45 minutes,the recovery and the coefficient of variation (CV)of the immunomagnetic beads adsorption method were 98.8% to 102.9% and 4.7% to 6.2%, respectively.The recovery and CV of the phosphate buffer saline method were 40.5%to 78.5%and 8.7%to 10.4%,respectively while the recovery and CV of the GB 29694—2013 method were 90% to 107.1%and 8.5% to 9.8%,respectively.The phosphate buffer saline method had low recovery(90%)and poor stability although it was rapid and convenient.Both GB 29694—2013 method and immunomagnetic beads adsorption method had high and stable recovery.The immunomagnetic beads adsorption method has advantage of time-saving and has remarkable potential for enriching target.
