甲氨蝶呤对类风湿性关节炎大鼠滑膜细胞凋亡基因调控的实验研究

Abstract
Objective:To observe the effective of M TX on RA synovial cell apoptosis and cell apoptosis gene in rats, and explore the mechanism of MTX on the disease.Methods:Rats were prepared for adjuvant arthritis models induced bacetic acid soluble type II collagen and Freund's complete adjuvant,and"MTX"as the intervention drug;The inhibitory effects on pawedana were observed.Separation of synovial cells in all groups rats and detected the expression of apoptosis relative protein by WB and FCM.Results:The result of experiment,the role of the experimental group were better than the control group(P<0.05);in both experiment,synovial cell apoptosis was increased in the model group.while Bax、Caspase-3 and Caspase-9 expression were increased,comparing with the control group and normal group,there were statis-tically significant(P<0.05).Conclusion:MTX can induce apoptosis of synovial cell,by down-regulating Bcl-2 expression and subsequently up-regulating Bax,Caspase-3 and Caspase-9 expression.关键词
