Abstract

Objective To cultivate the primary oligodendrocytes and examine JN expression at different stages of their differentiation and build eukaryotic expression recombinant plasmid contai-ning JN gene and explore its possible roles in the development of oligodendrocytes.Methods Oligo-dendrocytes were isolated and purified from cerebral cortex tissue of newborn rats and then the cells were identified against specific makers and JN by using Western blotting.The rat JN recombinant eu-karyotic expression plasmid was constructed by conventional method, including polymerase chain re-action(PCR), restriction enzyme digestion and ligation.The successfully reconstructed plasmid, as identified by sequence analysis, was tansfected into 293 T cells and detected by Western blottong.Meanwhile, the reconstructed plasmid was tansfected into OLN93 cells to observe the mor-phological changes of cells.Results Oligodendrocytes were cultivated successfully and JN was ex-pressed at the late stage of oligodendrocyte differentiation.Our results also showed that the JN recom-binant eukaryotic expression plasmid was successfully and constructed and JN could promote the for-mation of oligodendrocyte processes.Conclusion JN appeared at the late stage of oligodendrocyte development and it could promote the formation of oligodendrocyte processes and, possible, the for-mation of myelin sheath.
