机械压应力对人牙龈成纤维细胞表达钙网蛋白的影响
The effect of mechanical stress on the expression of calreticulin in human gingival fibroblasts

Abstract
Objective:To investigate the expression of calreticulin (CALR) in human gingival fibroblasts (HGFs) at different time under the action of mechanical pressure.Methods:HGFs were cultured by tissue block culture method.The cells were seeded on the PLGA scaffold,and then subjected to 25 g/cm2 loading force for 0 h (control),6 h,24 h,48 h and 72 h,respectively.The expressions of CALR mRNA and protein were detected by fluorescence quantitative PCR (qPCR) and western blotting,respectively.Results:The mRNA and protein expressions of CALR were up-regulated with the increasing of loading duration,and reached the highest level at 24 h,then decreased gradually.Conclusion:Mechanical compressive stress could up-regulate CALR expression in HGFs.The CALR level increased with prolonged duration of loading force,and reached the peak then decreased.关键词
