Abstract
Objective:To investigate the effect of mechano growth factor (MGF) on the proliferation of CD4+CD25+CD127-regulatoryT cells (Treg) in vitro.Methods:CD4+CD25+CD127 Treg cells in peripheral blood from healthy person were isolated by magnetic activated cell sorting (MACS).After purified,the cells were labeled with carboxyfluorescein (CFSE) and treated with different concentrations (0 nmol/L,10 nmol/L,100 nmol/L) of MGF.Treg cells proliferation was analyzed using flow cytometry.Peripheral blood mononuclear cells (PBMC) were isolated and treated with different concentrations (0 nmol/L,10 nmol/L,100 nmol/L) of MGF.The proportion of Treg cells in PBMC was detected by flow cytometry.Results:The cell proliferation rate was higher in 100 nmol/L MGF group than 0 nmol/L MGF group (P<0.05).The proportion of Treg cells in PBMC was 17.82%±1.23% in 0 nmol/L MGF groug,20.73%±1.61% in 10 nmol/L MGF group,and 20.29%±1.46% in 100 nmol/L MGF group.After treated with 10 nmol/L and 100 mol/L MGF,the proportion of Treg cells in PBMC was markedly increased,compared with 0 nmol/L MGF group (P <0.05),but no significant difference was noted between 10 nmol/L MGF group and 100 nmol/L MGF group (P>0.05).Conclusion:MGF could promote the proliferation of CD4+ CD25+ CD127-Treg cells in vitro.关键词
