摘要

Abstract

Objective:To study the regulation of TLR9-MYD88-TRAF6-IRF5 signaling pathway in systemic lupus erythematosus (SLE).Methods:38 SLE patients treated in our hospital from May 2016 to October 2017 were selected as an observation group,19 healthy subjects were enrolled as a control group in the same period.The level of interferon (IFN)-α in serum was detected by enzyme linked immunosorbent assay (ELISA).The mRNA expressions of Toll-like receptor 9 (TLR9),myeloid differentiation primary-response protein 88 (MYD88),tumor necrosis factor receptor-associated factor 6 (TRAF6) and interferon regulatory factor 5 (IRF5) in peripheral blood mononuclear cells (PBMC) were analyzed by qPCR.Results:The IFN-α level in serum and TLR9,MYD88,TRAF6 and IRF5 mRNA levels in PBMC were significantly higher in the observation group,compared with the control group (P <0.05).The TLR9 expression was positively correlated with the MYD88,TRAF6,IRF5 and IFN-α expressions as well as the SLE disease activity index,and was negatively related to complement C3 and complement C4 (P < 0.05).Conclusion:The TLR9-MyD88-TRAF6-IRF5 signaling pathway might play a role in the pathogenesis of SLE,the activation of which could result in active SLE.
