摘要
Abstract
To observe the pathological changes of retina and optic nerve of the optic nerve injury rats,and to explore the effect of Xuefu Zhuyu Decoction on the morphology and structure of the retina.Methods:Thirty-six rats were divided into normal group (n =4),treatment group (n =16) and control group(n =16).The rat optic nerve injury rat model was established in the treatment group and the control group,and 3 days,1 week,2 weeks,4 weeks after the model establishment,the slices were made,the changes of retinal morphology and optic nerve ultrastructure were observed by light microscope and electron microscope respectively.Results:① Retina:The 3rd day:Each layer of retina,especially the nerve fiber layer edema increased.The ganglion cells became swollen.The 1 st and 2nd week:The thickness of nerve fiber layer decreased with edema reducyion.A large number of ganglion cells decreased,and the inner nuclear cells decreased slightly.The inner plexiform layer and the outer plexiform layer edema increased.The 4th week:Each layer of retina edema reduced.The nerve fiber layer of Control Group was thinner than that of Treatmenit Group,and the ganglion cell density of Treatment Group was higher than that of Control Group.②Optic nerve:The 3 rd day:Lamellar separation of myelin can be seen.The axons became swollen and reduced.The numbers of microfilaments and microtubules were both reduced significantly.The 1 st week:Lamellar separation of myelin can be seen further expansion than before.Parts of nerve fibers degradated and formed "onion-like body".The 2nd week:The myelins bacame thiner.Some axons disappeared.The glial cells proliferated.The 4th week:The ultrastructures of optic nerve in the two groups were all repaired.Compared with Control Groups,the lamellar structure of myelin of Treatment Group repaired obviously.A large number of axons gradually restored,and the electron density increased.The numbers of microtubule,microfilament increased.The mitochondria structure was clear.The glial cells were almost normal.Conclusion:Retina and optic nerve can repair itself after trauma.Xuefu Zhuyu Decoction can reduce the retina and optic nerve structure damage.关键词
