丹参酮ⅡA对鼠新生血管的影响研究
Influence of Tanshinone HA in Neovascularization of Mice

Abstract
Objective:The study aimed to investigate the influence of the Tanshinone (TS) ⅡA in neovascularization of mice.Methods:Separately adopting the Matrigel implant method of b-FGF inducing mice abdominal wall and the way of culturing the rat aortic ring model of angiogenesis in vitro,the angiogenesis experiments were carried out from in vivo and in vitro.Then the hemoglobin content in the specimens was tested,the microvascular growth was observed under a microscope,the microvessel density was counted,the microvascular structural features and their developmental stages were observed.Results:(1) By detecting the mice Matrigel implants,we found that compared with the control group,the hemoglobin contents were lower in TS ⅡA high and low dose group and the endostatin group(P < 0.01).The microvascular count results were consistent with the hemoglobin content test results.(2) By culturing the rat abdominal aortic rings in vitro,we found compared with the controlled group,the microvascular budding area was decreased in 10 ~ 50% TS ⅡA serum group(P < 0.05,P < 0.01);the areas were increased in 70% and 80% TS ⅡA serum group(P < 0.01).The results of neonatal microvascular counts were consistent with the budding area.When the serum concentration was not more than 50%,the drug inhibition of vascular rings neovascularization levels was enhanced with their own concentration increased.At the same time,the vascular rings neonatal microvascular densities reduced gradually.Especially when the concentration was 50%,there was a significant difference(r =-0.563,P <0.01).Conclusion:TS ⅡA has an inhibitory effect on neovascularization and this inhibitory effect is concentration dependent.关键词
