Abstract

In order to understand the effects of homogentisate on the tyrosine degradation pathway of Arabidopsis thaliana and to explore whether the tyrosine degradation pathway in plants is inhibited under long-day conditions,the effects of homogentisate on the seeding growth under long-and short-day regimes were investigated using the materials from a wild-type A rabidopsis thaliana (Col-0) and two tyrosine pathway-deficient mutants (sscd1 and hgo).The effects of homogentisate on expressions of HGO,MAAI and SSCD1,the three key genes of tyrosine degradation pathway,were analyzed by qRT-PCR.The study showed that exogenous homogentisate accelerated the death of sscd1 mutant under short-day conditions,and also led to the death of sscd1 mutant under long-day conditions.Moreover,homogentisate promoted the expression of MAAI and HGO in both wild type and sscd1 mutant,and the MAAI expression level was higher in sscd1 than that in the wild type.Therefore,the above results indicated that exogenous homogentisate could activate tyrosine degradation pathway in plants and would not inhibit the pathway under long-day conditions.Further studies proved that the accumulation of intermediate metabolites in tyrosine degradation pathway would lead to the death of sscd1 mutant.
