DEHP对高脂饮食大鼠血脂代谢的影响
Effect of DEHP on the Lipid Metabolism in Rats Fed with High-Fat Diet

Abstract
Di-(2-ethylhexyl) phthalate (DEHP),known as an endocrine disrupting chemicals,is closely related to metabolic syndrome (MS).In order to investigate the relationship between DEHP and MS,an intragastrical administration method was adopted to evaluate the effect of DEHP on the lipid metabolism in rats fed with high-fat diet (HFD).Thirty male Wistar rats were randomly divided into 5 groups.The rats in control group were fed with regular chow,while rats of the other 4 groups were fed with high-fat chow and treated with different concentrations of DEHP (0,10,100 and 300 mg/kg body weight/day) for 5 weeks by gavage.At the end of the experiment,weight gain,Lee's index,organ/body weight ratio,the levels of serum lipids and adipokine were examined.The results showed that:(1) Body weight of rats wasn't significantly changed by DEHP (P>0.05).DEHP at the dose of 300 mg/kg resulted in the significant decrease in Lee's index and waist circumference,and markedly increased the organ/body weight ratio of liver or kidney (P<0.05).(2) The TG,TC and LDL-C levels of the rats (10 mg/kg DEHP group) were significantly elevated (P<0.05).The levels of TG and TC in 300 mg/kg DEHP group were reduced (P<0.05) and 100 mg/kg DEHP did not markedly influence the levels of TG and TC (P>0.05).(3) The level of leptin was not affected by DEHP,300 mg/kg DEHP notably increased the level of adiponectin compared with the HFD group (P<0.05).It suggests that DEHP influence lipid metabolism.DEHP at low dose might increase the risk of MS,the effect of DEHP at high dose needs a further study.关键词
