肢体缺血处理对缺血再灌注大鼠心肌PPARα mRNA和蛋白表达的影响
Impact of repetitive limb-ischemia conditioning on expressions of PPARα mRNA and protein in myocardium of rats following myocardial ischemia-reperfusion injury

Abstract
Objective:To observe the levels of CK-MB、cTnI and the expressions of peroxisome proliferator activated receptor-α(PPARα) mRNA and protein in myocardium of rats following myocardial ischemia-reperfusion(IR) injury and the impact of limb-ischemia (LI) conditioning for three times during different myocardial ischemic period.Methods:Fourty Wistar rats were randomly divided into 4 groups:IR group,LI-perC group (LI perconditioning),LI-preC group (LI precondioning),LI-postC group (LI postcondioning),ten rats in each group.Serum specimens and injured cardium of rats in four groups were kept after treatment.The contents of CK-MB and cTnI in the serum were detected according corresponding kits.The expressions of PPARα mRNA and protein in the myocardium were evaluated by reverse transcription polymerase chain reaction (RT-PCR) and Western blot respectively.Results:The levels of CK-MB and cTnI in LI-perC group,LI-preC group and LI-postC were significantly lower than those in IR group(P<0.05),while the expressions of PPARα mRNA and protein in the three groups were significantly higher than those in IR group (P<0.05).There were no significant changes in the contents of CK-MB and cTnI,the expressions of PPARα mRNA and protein in LI-perC group,LI-preC group and LI-postC group (P >0.05).Conclusion:.Limb-ischemia conditioning during different myocardial ischemic period may decrease myocardial injury and increase the expressions of PPARα mRNA and protein.关键词
