局部应用骨形态发生蛋白-7复合纤维蛋白治疗大鼠骨质疏松性椎体骨折的可行性
The feasibility of local application of BMP-7 fibrin complex in the treatment of osteoporotic vertebral fractures

Abstract
Objective To investigate the effect of bone morphogenetic protein 7(BMP-7) fibrin complex on osteopo-rotic vertebral fracture healing. Methods The experiment was done in animal experimental center of Xinjiang Medical Uni-versity from March 2017 to August 2017,72 female SD rats of clean grade were selected and randomly divided into experimen-tal group,model group and sham operation group;24 rats in each group. Both the experimental group and the model group were ovariectomized to establish the osteoporotic model, while the sham operated group was only excised some soft tissue a-round the ovary;After 3 months,three groups of rats were established fracture model,the experimental group was treated with 24 μg BMP-7+ fibrin 20 μg mixture to treat the fracture site,and the model group and the sham operation group were put into equal volume calf serum at the operation site. After operation,HE staining was used to observe the bone formation in the three groups;the vertebral mechanics related indexes of three groups were tested; the morphological parameters of vertebral body were measured in three groups of rats. Results (1)At fourth weeks after operation,the L5vertebral fracture of the experi-mental group and sham operation group showed a large number of cartilage cells and reticular fiber connection, and the L5 vertebral fracture of the model group showed a large number of cartilage cells and reticular fiber small connection. At eighth weeks after operation,both in the experimental group and the sham operation group had bony callus formed, and the fibrous callus disappeared.The callus formation in the model group was relatively loose,and there were a lot of fibrous callus. (2)At sixth,eighth,twelfth weeks after operation:the maximum load, the maximum strain, and the elastic modulus values of the model group were lower than those at the same period in the sham operation group and the experimental group,the differences were statistically significant(F=24.513,F=6.087,F=20.396,P=0.000,P=0.019,P=0.000);There was no statis-tical difference between the sham operation group and the experimental group (F =1.354, F =0.861, F =1.176, P =0.078,P=0.124,P=0.095). (3)At twelfth weeks after operation: TBV% and MTPT of rats in model group were lower than those at the same period in the sham operated group and the experimental group(F=4.589,F=2.079,P=0.031,P=0.046),TRS% and MAR were higher than those in the sham group and the experimental group(F=4.589,F=2.079,P=0.031,P=0.046). There was no significant difference between the experimental group and the sham operation group (F=0.643,F=0.652,F=1.431,F=0.038,P=0.165,P=0.161,P=0.074,P=1.049). Conclusion Focal application of BMP 7 fibrin complex has significant promoting effect on the healing of osteoporotic vertebral fracture in rats.关键词
