蛋鸡Ⅰ群腺病毒湖北株的分离与分子鉴定
ISOLATIONS AND CHARACTERIZATION OF GROUP Ⅰ ADENOVIRUS FROM SICK LAYING HEN IN HUBEI PROVINCE

Abstract
Liver samples were collected from layers suspected "pericardial effusion and hepatitis syndrome" on different farms. The virus was isolated from the samples and inoculated with SPF chicken embryos after treatment. The viral DNA was extracted and amplified in PCR for sequencing. Subsequently, 30-day-old healthy chicks were inoculated with the isolated virus and clinical signs and pathological changes were observed. Three avian adenoviruses were isolated with 100% gene homology. These isolates shared 99.8% similarity of nucleotide sequences to those of serotype 4 of adenovirus group I and 43.1%-78.6% to those of adenovirus groups Ⅱ and III. Some of inoculated chicks died at day 3 post inoculation and the number of deaths peaked at day 5. Total chick mortality reached to 60% during the observation period. The typical signs of pericardial effusion and inclusion body hepatitis were observed. In addition, fatty and vacuolar degenerations were found in liver cells and vacuoles and necrosis in kidney cells. The results indicated that group Ⅰ Adenovirus was isolated from sick laying hen, named as Hubei strain.关键词
