帕金森病模型大鼠脑内CDNF表达变化
Expression of CDNF in the brain of the parkinson's disease rat
摘要
Abstract
In order to investigate the expression of CDNF in the process of Parkinson's disease (PD),we used rat as an experimental model to determine the expression of dopaminergic neurotrophic factor and its related genes in the brain of PD to to provide a theoretical basis for gene therapy of PD.The rat model of PD was prepared by single-point injection of 6-hydroxydopamine in unilateral substantia nigra.Immunohistochemistry,western blot and Q-PCR methods were used to detect the expression of CDNF and threonine-hydroxylase (TH) in the substantia nigra of the model rats.We found that the number of CDNF and TH positive cells in the substantia nigra of the model rats was significantly decreased.Further research showed that when compared with the normal control rats,CDNF mRNA expression showed no change in the model rats (P>0.05),while CDNF and TH protein expression were significantly reduced (P<0.01).These results suggested that the incidence of PD may be relevant to the decreased expression of CDNF and TH in the substantia nigra,which further inhibit dopamine synthesis.关键词
