Abstract

The RWP-RK gene family is a plant-specific transcription factor family.RWP-RK transcription factors play central roles in regulation of plant nitrogen signal transduction and gametophyte development.Using the genomic sequences of "Capsicum annuum L.Zunla-1",an important pepper cultivar in China and its ancestor Mexican pepper "Chiltepin",we identified the genes containing the RWP-RK domain in these two pepper genomes.We also carried out phylogenetic,chromosome localization,conserved sequences and possible function analysis.In the genome of Capsicum annuum L.Zunla-1,CaZunla01g000097,CaZunla01g004485 and CaZunla00g000405 were homologous to the essential regulatory factor (AtNLP7) controlling the nitrogen metabolism in Arabidopsis thaliana.Capana03g001990 and Capana03g000787 were homologous to Arabidopsis thaliana egg formation related transcription factors AtRKD1,AtRKD2 and AtRKD4.These genes maintain a high degree of conservation between the cultivar Zunla-1 and its wild ancestor "Chiltepin".In addition,there is a unique RWP-RK transcription factor subfamily in the pepper genomes.This subfamily protein contains unique motifs that are not existed in the Arabidopsis RWP-RK transcription factors.There is also a RWP-RK transcription factor unique in Capsicum annuum L.Zunla-1,but not in the wild ancestor "Chiltepin".In this study,we identified RWP-RK transcription factors that are highly similar or different to Arabidopsis RWP-RK transcription factors.These identified factors showed significant differences between the wild and cultivars of pepper.The results provide important information for pepper RWP-RK genes cloning and functional analysis.Functional studies on these genes will greatly promote the study of the molecular mechanism of peppers in response to nitrogen deficiency and pepper gametophyte development.
