摘要

Abstract

Objective To study the interaction between bovine serum albumin (BSA) and inosine and effects of metal ions.Methods Under the optimal conditions,the interaction between BSA and inosine and effects of metal ions were studied by UV-visible absorption spectra and fluorescence spectra.The quenching constant (KSv),speed constant (Kq),static quenching constant (KLB),binding constants (Kb),the number of binding site (n),enthalpy change (△H),entropy change (△S),free energy change (△G) and Hill' s coefficients (nH) were analyzed according to Stem-Volmer equation,double reciprocal equation,thermodynamic equation and Hill equation.Results Inosine could quench the fluorescence of BSA,which was a static quenching process.BSA could transport inosine.The hydrogen bonds and Van der Waals forces played an important role in the conjugation reaction of BSA and inosine.The primary binding site for inosine was located at sub-domain Ⅱ A of BSA and nearby tyrosine residue.There was negative weak cooperative effect.The conjugation reaction would affect the conformation of BSA.Mn2+,Co2+,Cu2+ and Ni2+ played a competitive role in the binding of inosine to BSA,and enhanced the efficacy.Cr3 + promoted the interaction and prolonged drug effect time.Conclusion This study provided a theoretical basis for revealing the pharmacokinetics and clinical application of new coenzyme drugs.
