Abstract

The genetic diversity and relationship as well as maternal origin among them of 179 individuals from Xianan cattle population and 13 other cattle populations were analyzed by the detection of polymorphisms in mitochondrial D-loop sequences and genome microsatellite DNA.Sequencing and analyzing D-loop region of 179 individuals belonging to 14 cattle populations found 124 variable sites and 60 haplotypes.So,it indicated that 14 cattle populations had rich genetic diversity.There were 49 variable sites and 7 haplotypes in mtDNA D-loop region sequence of Xianan cattle individuals,the haplotype diversity was 0.732±0.017 and its haplotype diversity was lower than other Chinese cattle.Chinese cattle had richer genetic diversity.Sequencing and analyzing the allele numbers,mean heterozygosity (H) and polymorphism information content (PIC) values of the nine high polymorphic microsatellite loci showed that 47 alleles were found at 9 microsatellite loci in 60 Xianan individuals and the average number of alleles per locus ranged from 6.584 to 1.229.The polymorphism information and average heterozygosity content of 14 cattle populations ranged from 0.608 ~0.798 and 0.695 ~0.837,while they were 0.692 and 0.715 for Xianan cattle population.These results suggested that genetic diversity of Chinese cattle was slightly higher than Xianan cattle population and exotic cattle breeds,which were consistent with the studied results of mtDNA Dloop region among populations.NJ (neighbor joining) tree showed that 14 cattle populations mainly originated from bos taurus and bos indicus,then Xianan cattle population with the Germplasm Characteristics of Bos indicus and Bos taurus was greater affected by bos indicus.This research offers theoretical basis for protection,exploitation and breeding improvement of the genetic resources in Xianan cattle population.
