miR-137通过TCF-4抑制胶质瘤细胞生长侵袭能力的体外研究
Suppressive effect of miR-137 on invasive ability of glioma cell through TCF-4 in vitro

Abstract
Objective To explore the effect and mechanism of miR-137 on invasive ability of LN229 glioma cells.Methods The miR-137 mimics were transfected to human glioma cell line LN229.The expression of miR-137 was identified by Quantitative real-time PCR(qRT-PCR)after transfection.Methylthiazol tetrazoliu(MTT),transwell assay was employed to detect the cell invasive ability.Relative targeting protein expression was assayed by Western blotting.Results After transfection of miR-137 mimics,the miR-137 mimics treated cells survival rate was decreased obviously in comparison with blank group cells by the MTT assay.miR-137 mimics had greatly suppressed ability to invade matrigel in LN 229 cell,exhibiting about 44.4 ±6.8,42.2 ±6.1,16.9 ±2.1 respectively in blank group,negative control group and mimics treated group cells.Moreover, Tcf-4 protein expression level were inhibited after transfected by miR-137 mimics.Conclusion miR-137 inhibits proliferation and invasive ability of glioma cells,suggesting that miR-137 could be tumor suppressor gene in glioma.关键词
