摘要

Abstract

Objective To investigate the inhibitory effect and its mechanism of Lutein on myocardial fibrosis (MF) in rats.Methods Wistar rats were randomly divided into five groups according to the weight:normal group,model group,high,middle and low three doses of the experimental group (20,40,80 mg · kg-1 · d-1 Lutein),10 rats in each group.Except normal group,the rats in other groups were used to construct MF models by subcutaneous injection of isoproterenol (Iso:5 mg · kg-1 · d-1) for 7 days,and experimental groups were given different doses of Lutein from second days.Normal and model groups were given the same amount of distilled water.After the end of the experiment,heart mass index (HMI) and the left ventricular mass index (LVMI) were measured by analytical balance.Procollagen I C-terminal propeptide (PICP) and procollagen Ⅲ N-terminal propeptide (PmNP) were detected by ELISA method.The content of serum lactate dehydrogenase (LDH) and creatine kinase (CK) and nitric oxide(NO) level were detected by microplate reader.The activity of calcineurin (CaN)and the content of malondialdehyde (MDA) in myocardial tissue of rats were measured by ultraviolet spectra.Results Compared with normal group,in model group:HMI (mg · g-1) and LVMI (mg · g-1) were 3.73 ±0.58 and 2.57 ±0.43;PICP(ng · mL-1) and PⅢNP(ng · mL-1) were 7.27 ±0.33 and 24.86 ± 1.88,LDH and CK were 465.42 ±47.05 and 489.21 ±27.70;NO(μmol · L-1)was 2.06 ±0.34;MDA (nmol ·mg-1) content was 2.93 ±0.51 and the activity of CaN (U ·mg-1) was 0.14 ±0.02,the difference of above data between two groups were statistically significant (P <0.05 or P <0.01).Compared with the model group,experimental-L,-M,-H groups(unit:refer model group):HMI and LVMI were 3.13 ±0.40,3.02 ±0.28,2.82 ±0.22 and 2.19 ±0.27,2.09 ±0.27,1.99 ±0.12;the content of serum PICP and PⅢNP were 6.44 ±0.69,6.23 ±0.29,6.14 ±0.75 and 22.17 ±2.10,21.88 ± 1.93,21.76 ± 1.09;the content of LDH and CK in serum were 442.13 ± 33.72,390.19 ± 41.31,344.89 ± 50.25 and 442.89 ± 37.40,369.30 ± 28.46,346.57 ± 28.56;the level of NO in serum were 3.07 ± 0.45,4.59 ± 0.64,5.16 ± 0.52;the content of MDA and activity of CaN were 0.07 ± 0.01,0.06 ± 0.00,0.04 ± 0.01 and 2.48 ±0.27,2.44 ±0.17,2.25 ±0.11;these data were obviously significant (P <0.05 or P <0.01).Conclusion Lutein can be antioxidant and prevent calcium channel function by increasing the level of NO,reducing the content of MDA and the activity of the CaN to inhibit the synthesis of collagen and the myocardial fibrosis of rats to protect the myocardium.
