摘要

Abstract

Objective To observe the expression of fibroblast growth factor 2 (FGF-2),epidermal growth factor (EGF),monocyte chemotactic protein 1 (MCP-1) and stromal cell derived factor 1 (SDF-1)in calvarial bone defect rat,explore the mechanism of Musk promote bone defect healing.Methods Cranial bone defect model was prepared with cranium drill.Sprague-Dawley rats were randomly divided into model group and experimental group,150 rats in each group.The rats in the experimental group were given 42 mg · kg-1 of natural Musk,the model group were given the same amount of normal saline,1 times a day.The expression of FGF-2,EGF and MCP-1 was determined by Western blotting and SDF-1 was analyzed by ELISA method on 7,14 d and 28 d respectively after administration.Results After administration,the relative expression levels of FGF-2 in model group and experimental group on 7,14 d were 0.17 ± 0.00,0.41 ± 0.03;0.10 ± 0.01,0.68 ± 0.05,respectively.Compared with model group and experimental group,the difference was significant on 7.d and 14 d (all P < 0.05).The relative expression levels of EGF in model group and experimental group at 7,14 d were 0.12 ±0.01,0.16 ±0.02;0.08 ±0.01,0.45 ±0.03,respectively.Compared with model group and experimental group,the di-fference was significant at 7,14 d (all P < 0.05).The relative expression levels of SDF-1 in model group and experimental group on 7 d,14 d were (63.33 ± 8.09),(76.95 ± 6.82);(70.56 ± 11.91),(71.80 ± 8.50) ng · mL-1,respectively.Compared with model group and experimental group,the difference was significant at 7,14 d (all P < 0.05).Conclusion Musk can promote the healing of cranial bone defect in rats,and its mechanism may be related to the chages of expression of FGF-2,EGF,MCP-1 and SDF-1.
