摘要

Abstract

Objective:To preliminarily explore the effect of nitidine chloride (NC) on apoptosis of HT29 colorectal cancer cell line and the related mechanism.Methods:HT29 cells were divided into vehicle control group and NC groups (15,30 and 60 μmol/L).CCK-8 assay and flow cytometry were used to detect cell proliferation and apoptosis,respectively.Western blotting was employed to determine the protein levels of cytochrome-c (Cyt-c) and caspase-3.Results:Compared with the vehicle control group,the cell viability rates of the NC groups decreased obviously and the apoptosis rates [(35.30±14.30)%,(41.48±17.89)% and (46.51 ± 11.35) %] increased prominently,and differences were statistically significant (P <0.05).Western blot showed that the protein levels of Cyt-c and caspase-3 in the NC groups were notably upregulated compared with the vehicle control group (P<0.05).Conclusion:NC could significantly inhibit the proliferation of HT29 cells and induce apoptosis,and the mechanism might be associated with the Cyt-c mediated mitochondrial apoptotic pathways.
