Abstract
Objective:To study the effects of hydroxyurea on γglobin gene expression in K562 cells and the regulation of microRNA that may be involved.Methods:K562 cells were treated with different concentrations of hydroxyurea (0 μmol/L,100 μmol/L,200 μmol/L,300 μmol/L,and 400 μmol/L) for 24 h,48 h,72 h and 96 h.Real-time fluorescent quantitative PCR (qPCR) was used to detect the expression of γ globin and relevant microRNA.Results:The expression of γ globin gene,miR-451a and miR-503-3p in K562 cells were significant increased in the hydroxyurea groups,compared with the control group (P<0.05 or P <0.01).Conclusion:Hydroxyurea could up-regulate γ globin gene expression in K562 cells,probably though the modulation of miR-451a and miR-503-3p expressions.关键词
