多西环素对大鼠烟曲霉菌性角膜炎中炎症细胞及MCP-1的抑制作用
Inhibitory effect of doxycycline on the inflammatory cells and the expression of MCP-1 in rats with Aspergillus fumigatus keratitis

Abstract
Objective:To investigate the effect of doxycycline on the inflammatory cells and the expression of monocyte chemoattractant protein-1 (MCP-1) in rats with Aspergillus fumigatus keratitis (AFK).Methods:Sixty-four SD rats were randomly divided into a control group and a doxycycline group,with 32 rats in each group.All rats were established the AFK model.Rats in the doxycycline group were treated with 0.3 g/L doxycycline eye-drops,and those in the control group received menstruum eye-drops.Corneal inflammatory index was recorded on the 3rd,5th,7th,and 10th day.The mRNA and protein levels of MCP-1 were detected by fluorescence quantitative PCR (qPCR) and enzyme-linked immunosorbent assay (ELISA),respectively.Results:Compared with the control group,the inflammatory index was decreased in the doxycycline group on the 3rd,5th,and 7th day,and the inflammatory cell count was reduced at any time points (P<0.05).The expression of MCP-1 mRNA and protein was down-regulated on the 3rd,5th and 7th day after doxycycline treatment (P<0.05).Conclusion:Doxycycline could decrease the infiltration of inflammatory cells in rats with AFK,the mechanism might be related to the inhibition of MCP-1 expression.关键词
