摘要

Abstract

Objective:To investigate the mRNA expressions of NF-κB p65,IκBα and IL-1β in radiation-induced lung injury rats.Methods:64 male Sprague-Dawley rats were randomly divided into a control group and an experimental group.Rats in the experimental group received 18 Gy irradiation to induce lung injury.After 2,6,12 and 24 weeks,the lung tissue was excised and processed for hematoxylin-eosin (HE) staining and Masson staining.Hydroxyproline was measured using a method based on alkaline hydrolysis.The interleukin-1β (IL-1β) level in serum was examined by enzyme-linked immunosorbent assay (ELISA).The mRNA expressions of NF-κB p65,IκBα and IL-1β were determined by fluorescent quantitative PCR (qPCR).Results:Alveolar structure of lung tissue in the control group was intact without edema and exuda tion.Pathological changes in the lung tissue of all irradiated rats exhibited early hyperemia,edema,collagen deposition,inflammatory cell infiltration,thickened alveolar wall and alveolar septum,then aggravated pulmonary fibrosis and smaller alveolar space.The content of hydroxyproline was higher in the experimental group than that in the control group at 6,12 and 24 after radiation (P<0.05).Serum IL-1β level and the expressions of NF-κB p65 and IL-1β were increased,while the IκBα mRNA level was decreased in the experimental group at each time point,compared with the control group (P <0.05).IL-1β content in serum and NF-κB p65 and IL-1β mRNA levels in lung tissue reached the pick at week 6 of radiation,then declined gradually,but the IκBα mRNA level showed the opposite trend.Conclusion:The mRNA expressions of NF-κB p65 and IL-1β were notably up-regulated and IκBα was down-regulated at the early stage of radiation-induced lung injury in rats.Inhibition of NF-κB p65 and inflammatory response might attenuate radiation-induced lung injury.
