两种小鼠颈总动脉内膜增生模型的制备和比较
Construction and comparison of two mouse models of common carotid arterial neointimal hyperplasia

Abstract
Objective:To construct two mouse models of common carotid arterial neointimal hyperplasia (common carotid artery ligation model and common carotid wire injury model) and compare the advantages and disadvantages of these two models.Methods:Ligation or wire injury operation were performed on C57BL/6 mice in the right common carotid artery,leaving the left side as a control.Fourteen days after the operation,common carotid artery ultrasonography was performed to observe the blood flow velocity (BFV) in the carotid artery.Hematoxylin-eosin staining was performed to observe intima areas,media areas and intima to media ratios.Immunofluorescence of SM22 staining was performed to observe the degree of dedifferentiation of vascular smooth muscle cell (VSMC).Results:The blood flow disappeared in the ligated side but accelerated in the opposite side in comparison with the normal carotid artery.The BFV in both the wire injury sides and the opposite sides accelerated compared with the normal ones with the former being faster.The intima areas of ligation sides and injury sides was (35.08± 4.40) × 103 μm2 and (12.02±2.00) × 103 μm2 respectively;the media areas was (49.31± 3.39) ×103 μm2 and (39.63±4.05)× 103 μm2 respectively;intima to media ratios was (0.71±0.05) and (0.30±0.05)respectively.All differences had statistical significance.The VSMC in the operative side switched from differentiated state to dedifferentiated state with a higher dedifferentiated degree in the ligation sides.Conclusion:In the respect of imitating arterial neointimal hyperplasia,common carotid artery ligation model was superior to wire injury model.关键词
