Abstract

Objective:To investigate the expression of β-catenin and CD146 in mouse endometrium during regeneration.Methods:Peanut oil was injected into the uterine lumen followed by ovariectomy (OVX) 48 hours later to establish endometrial regeneration model.The uterine horn was harvested at 0,24,48,72 h and 96 h after OVX respectively,and then the expression of active β-catenin and CD146 protein was detected by immunohistochemical staining.Meanwhile,β-catenin mRNA level was estimated by qRT-PCR.Results:Decidual reaction appeared extensively in the endometrium at 0 h after OVX,followed by necrosis and isolation of the decidual zone at 24 h.The endometrium was thickened obviously at 48 h,and then the regeneration was completed at 96 h.Immunohistochemical analysis showed that positive staining of active β-catenin in the endometrium was very weak at 0 h after OVX,and increased subsequently after 24 hours,followed by a peak at 48 h.Nevertheless,the positive intensity began to decline at 72 h,continuing to fall at 96 h.The pattern of CD146 showed some similarities to active β-catenin.At 0 h after OVX it was mainly distributed in the vascular endothelium and seldom in the stroma cells.The total intensity slightly decreased at 24 h but it increased in some stroma cells,significantly reached a peak at 48 h,and then began to decline at 72 h,continuing to fall at 96 h.The qRT-PCR results showed that the relative quantity of β-catenin mRNA increased slightly at 48 h (1.63±0.13) and reached a peak at 72 h (2.16±0.18),and then declined at 96 h (1.98±0.23).Conclusion:β-catenin and CD146 express in specific temporal and spatial patterns in the repair process of the endometrium,implying that Wnt/β-catenin signal pathway is involved in endometrial repair process,and may be related to CD146+ cells which probably are candidates of potential endometrial mesenchymal stem cells.
