摘要

Abstract

Objective:To investigate the effect of microencapsulated and non-microencapsulated olfactory ensheathing cells (OECs) transplantation on peripheral nerve injury and the expression of P2X3 purine receptor.Methods:NASH method was used to cultivate and expand cells from olfactory bulb tissue of a SD rat.SD rats were randomly divided into sham group,model group,non-microencapsulater-OECs (non-MC-OECs group) and microencapsulated OECs group (MC-OECs groups).The 10th and 20th days after surgery,the mechanical stimulation constraining thresholds of the rats in each group were measured by behavioral methods.P2X3 receptor positive cells percentage and average absorbance were observed by in situ hybridization histochemistry.Results:The 10th and 20th days after surgery,the mechanical stimulation constraining thresholds of rats were increased from the model group,the non-MC-OECs group,the MC-OECs group to the sham group,and the positive cells percentage and average absorbance of P2X3 receptor were decreased in turn.Compared with CCI group,the mechanical stimulation constraining thresholds of rats were increased,and the positive cells percentage and average absorbance of P2X3 receptor were decreased in the non-MC-OECs group.Compared with non-MC-OECs group,the mechanical stimulation constraining thresholds of rats were increased,the positive cells percentage and average absorbance of P2X3 receptor were decreased in the MC-OECs group.The difference between groups was statistically significant.Conclusion:Microencapsulated OECs transplantation can better alleviate chronic peripheral nerve injury pain and reduce the expression level of P2X3 receptor than non-MC-OECs on the 20th day after surgery.
