过表达整合素-α5促进牙髓干细胞顺硬介质表面迁移和向牙本质分化的研究
Overexpression of integrin-α5 enhances migration and differentiation to dentin of dental pulp stem cells on stiff substrates surface

Abstract
Objective:To promote the migration and differentiation to dentin of dental pulp stem cells(DPSCs)on stiff substrates surface by over expression of integrin-α5(ITGA5).Methods:After obtain of mouse DPSCs,its mesenchymal cell surface markers were detected by flow cytometry.And after lentiviral plasmid containing ITGA5 gene infection,the mRNA level of ITGA5 in DPSCs was tested by qRT-PCR method.Then DPSCs were cultured on the surface of agarose medium with the elastic modulus of 75~100 kpa.The migration ability was detected by cell scratch assay,and the cell invasion ability of a whole mass was detected by 2D-cultured assay.Then mineralized nodules were detected by alizarin red staining,and the pro-tein expression of dentin matrix protein 1(DMPl),dentin sialoprotein(DSP)and dentin phosphoprotein(DPP)was tested by Western Blot method.Results:The results of flow cytometry showed that the cultured cells express mesenchymal cell surface markers.After gene infection,the mRNA of ITGA5 could be stably expressed in DPSCs at least 30 days,and their migration and invasion ability was increased significantly,while more larger mineralized nodules were formed,with obvious high expres-sion of DMPl,DSP and DPP protein.Conclusion:Overexpression of ITGA5 can promote migration and differentiation ability to the dentin of DPSCs on the surface of stiff media,which can provide a new method for dental tissue engineering.关键词
