Abstract

Objective:To prepare dual factor-loaded scaffolds for controlled release of platelet-derived growth factor (PDGF)and simvastatin(SMV)and to evaluate the release properties of this formulation.Methods:SMV was encapsulated in collagen/polycaprolactone membranes using the electrospining method and surface wettability of the membranes was deter-mined by static contact angle measurement.Then,PDGF was assembled onto the surface of electrospun membranes using the electrostatic layer-by-layer coating method.The in vitro release profiles of PDGF and SMV were determined by an enzyme linked immunosorbent assay(ELISA)kit and an ultraviolet-visible spectrometer,respectively. Results:Reduction of water contact angle showed good hydrophilicity of the polycaprolactone membrane,which increased in the presence of collagen.The in vitro release analysis demonstrated a sequential release profile,which was characterized by the fast release of PDGF with sustained concentration from day-3 to day-13,followed by the slow release of SMV with sustained concentration from day-6 to day-21.Conclusion:Dual factor-released membrane scaffolds were successfully fabricated by encapsulating PDGF and SMV with distinct shell-core structure.The combination and sequential release of PDGF and SMV could be application in periodon-tal tissue engineering.
