多酚与淀粉样纤维相互作用新说
A new paradigm in polyphenols-amyloid fibrils interactions

Abstract
It was found in previous studies that one of the most impressive properties of polyphenols is the inhibition of amyloid protein fibril formation. In addition,they remodel or cut mature amyloid fibrils under low protein concentrations. We found for the first time that polyphenols-binding amyloid fibrils in the nematic phase(high protein concentration)induce controlled fibrils aggregation to form complex hierarchical structures with water-rich nature,so-called hydrogels without phase separation,which are mainly driven by the exogenous complementary hydrogen bonding,π-π stacking and hydrophobic interactions. Changing the type of amyloid does not significantly change the conclusions of the structure and properties of the hydrogels,which indicates that the effect of polyphenols on controlling gelation of amyloid fibrils is not protein-dependent. The ensued hydrogels exhibit strong antibacterial activities against both gram-negative and gram-positive bacteria,controlled by bacterial agglomeration,without any notable cytotoxicity to human cells. The anti-bacteria mechanism of the amyloid fibril hydrogels is initiated by membrane disintegration. The cationic, hydrophobic and conformational properties of the amyloid fibrils could be the main sources for the observed antibacterial activity.关键词
