Abstract

Objective To explore the effect of silent CACUL1 expression on the proliferation of esophageal cancer cells and its mechanism.Methods Real-time quantitative PCR was used to detect the change of CACUL1 mRNA expression after transfection CACUL1 siRNA in EC109 cells.EC109 cell proliferation was determined by MTT assay.Cell cycle and apoptotic rate were analyzed by flow cytometry(FCM),and the expression of CDK2-Cyclin A/E protein was detected by Western blot method.Results CACUL1 mRNA and protein expression significantly decreased after transfection with CACUL1 siRNA(P < 0.01).MTT test results showed that EC109 cell proliferation was inhibited significantly after transfection with CACUL1 siRNA(P <0.01).FCM results showed that the cell cycle was arrested in G1/G0 after transfection with CACUL1 siRNA(P <0.01),and that the early apoptosis and late apoptosis rate were increased significantly(P < 0.01).Western blot result showed that the CDK2-Cyclin A/E protein expression was down-regulated significantly after transfection with CACUL1 siRNA.Conclusion CACUL1 may promote the proliferation of esophageal carcinoma EC109 cells through regulating CDK2-Cyclin A/E signaling pathway.
