补骨脂酚对口腔鳞状细胞癌增殖、迁移和凋亡的影响及其机制
Effects of bakuchiol on proliferation, migration and apoptosis of oral squamous cell carcinoma and its underlying mechanisms

Abstract
Objective To elucidate whether silent information regulator 1 (SIRT1) signaling pathway mediates the effects of bakuchiol (BAK) on the proliferation,migration and apoptosis of oral squamous cell carcinoma(OSCC).Methods The OSCC cells were divided into four groups:control group,BAK group,BAK + EX527 (a selective SIRT1 inhibitor) group,and EX527 group.The effects of EX527 and different doses of BAK(1,2,5 and 10 μmol/L) on the cell viability of OSCC cells were determined by CCK-8 essay,the apoptotic ratio was determined by TUNEL staining,the expression levels of SIRT1,Ac-FOXO1,Bax,Bcl-2 and Caspase-3 were determined by Western blot,and the migration capacity of OSCC cells was measured using scratch assay.Results Compared with the control,BAK dose-dependently decreased the cell viability of OSCC cells,with the best effects at the dose of 5 μmol/L.Thus,5 μmol/L was used for the mechanism research.Compared with control group,the expression levels of SIRT1,Bax and Caspase-3,the apop-totic ratio of OSCC cells and the distance between the cell boundaries were significantly increased in BAK group,while the expression levels of Ac-FOXO1 and Bcl-2 were significantly decreased.However,these effects of BAK mentioned above were significantly decreased by EX527 (all P < O.05).Conclusion BAK may inhibit the proliferation and migration,and promote the apoptosis of OSCC cells via activating the SIRT1 signaling pathway.关键词
