摘要

Abstract

Objective To analyze the effect of Lycium barbarum polysaccharides on heat shock protein 27 (HSP27) expression and oxidative stress products in lens epithelial cells of diabetic cataract rats.Methods Healthy SD rats were selected and diabetic cataract rat models were established by intraperitoneal injection of streptozotocin.As the control group,another 15 SD healthy rats were given the same amount of 1 mol · L-1 sodium citrate buffer.The model rats were randomly divided into model group,test A and B groups with 16 rats per group.Test A group was treated with 12.5 mg · mL-1 hydrochloride solution,each with 2 mL,qd,intragastric administration.Test B group was treated with 25 mg · mL-1 Lycium barbarum polysaccharide solution,each with 2 mL,qd,intragastric administration.Model group and control group were both treated with sterilized 0.9%NaCl,each with 2 mL,qd,intragastric administration.4 groups of rats were treated for 12 weeks.The body weight,blood glucose,relative gray value of lens opacity,levels of oxidative stress products indexes and expression level of HSP27 protein in rats of the 4 groups were compared.Results After treatment,the main indexes in control,model,test A and B groups were compared:weight were(285.64 ± 19.54),(251.73 ± 10.85),(260.89 ± 11.47)and (270.45 ± 13.11)g,blood glucose were (8.12 ±0.83),(26.47 ±3.25),(15.18 ± 1.84) and (10.02±0.43) mmo · L-1 relative gray value of lens opacity were (20.28 ± 2.19) (85.66 ± 8.67) (51.18 ±6.77) and (42.24±5.14),malondialdehyde were (3.95 ±0.47),(8.15 ±0.84),(5.73 ±0.64) and (4.86±0.51) nmol · mg prot-1,superoxide dismutase were (75.74 ±7.64),(33.26 ±3.18),(54.07 ±5.72) and(62.78±8.36) U·mgprot-1,glutathioneperoxidasewere (15.78±1.85),(6.25±0.68),(9.88±1.29) and (13.02 ± 1.14) U · mg prot-1,HSP27 were (0.47 ±0.05),(0.73 ±0.09),(1.35 ±0.16) and (1.78 ±0.22).Compared with control group,those indexes had statistically significant differences in model group,test A and B groups (all P < 0.05).Compared with test A group,the indexes above of test B group had statistically significant differences(all P < 0.05).Conclusion Lycium barbarum polysaccharide maybe reduce the lens opacity and improve the situations of weight loss and hyperglycemia in diabetic cataract rats,by improving the antioxidant activity of the lens and promoting the expression of HSP27 in the lens epithelial cells.
