Effect of trichosanthin on the apoptosis and the expression of related gene in human breast cancer MCF-7 cell

Abstract
Objective To explore the effect of trichosanthin on the apoptosis and the expression of related gene in human breast cancer MCF -7 cell.Methods Logarithmic phase human breast cancer MCF-7 cell managed by 30,60,90,120 μg · mL-1 trichosanthin and acted as test group,control group managed by equivalent 0.9% NaCl,cultured for 24,48,72 h.The changes of cell morphology and nucleus apoptosis morphology were compared,the proliferation inhibition of human breast cancer MCF-7 cell was detected by methyl thiazolyl tetrazolium (MTT),the effect of trichosanthin on the expression of cysteinyl aspartate specific proteinase(caspase)-3 and caspase-8 of human breast cancer MCF-7 cell was deteted by spectrophotometry.Results The cell morphology and nucleus apoptosis morphology of human breast cancer MCF-7 cell in test group was obvious;and the changes of nucleus apoptosis morphology were more obvious with the increasing of the trichosanthin action concentration.The proliferation inhibition rates in 30,60,90,120 μg · mL-1test group at 24 h after managed were (1.61 ±0.94) %,(2.81 ±1.01) %,(7.05 ±1.03) %,(9.59±1.12)%.The proliferation inhibition rates at 48 h after managed were (2.13 t± 1.01) %,(6.14 ± 1.12) %,(9.05 ± 1.09) %,(19.60 ± 1.15) %.The proliferation inhibition rates at 72 h after managed were (3.28 ± 1.07) %,(7.18 ± 1.51) %,(18.39 ± 1.81)%,(29.59 ± 1.63) %.The proliferation inhibition rates in test group increased with the increasing of the trichosanthin action concentration and the extension of trichosanthin action time (P <0.01).The caspase-3 of 90 μg · mL-1 test group at 24,48,72 h were 1.49 ±0.15,2.36 ±0.25,2.15 ±0.23;and the caspase-8 were 1.94 ±0.26,1.56 ± 0.19,1.39 ± 0.20.The caspase-3 and caspase-8 of human breast cancer MCF-7 cell in control group had no significant changes with the extension of trichosanthin action time (P > 0.05),and the caspase-3 in 90 μg · mL-1 test group increased at first and then decreased (P <0.05 or P<0.01),and which at 48 h was highest.The caspase-8 decreased gradually (P < 0.01);the caspase-3 and caspase-8 in 90 μg · mL-1 test group at each time were higher than that in control group (P < 0.01).Conclusion Trichosanthin can promote the apoptosis of human breast cancer MCF-7 cell effectively and inhibit the proliferation,and proliferation inhibition is time-dose dependent,the action mechanism maybe related to the up-regulation of the expression of caspase-3 and caspase-8 in human breast cancer MCF-7 cell.关键词
