Abstract
Objective To investigate the effect of miRNA-769-5p on biological activity in normal human fibroblasts. Methods Human fibroblasts were transfected with miRNA-769-5p mimic. CCK-8 kit was performed to test proliferation rate. Cell were observed under fluorescence microscope. Flow cytometer was used to measured rate of reactive oxygen species and apoptosis. All the parameters of the mimics group were compared with the control group. Results The relative expression of miRNA-769-5p in the mimics group was (1.36±0.2), which was significantly higher than that of the control group (1.0±0.1), the difference was statistically significant(P<0.05). The cell proliferation rate in the mimics group (0.747±0.072) was significantly lower than that of the control group (1.007±0.025)(P<0.05). The fluorescence value of the mimics group (92.067±3.092) was significantly higher than that of the control group (21.600±8.487)(P<0.05). The apoptotic rate in the mimics group was (49.299 ± 5.184)%, which was significantly higher than that of the control group (5.977±0.939)%(P<0.05). Conclusion Up-regulation of miRNA-769-5p induce photo-damage and have effect on biological activity in normal human fibroblasts.关键词
