大肠癌中miR-10b和HOXD10mRNA的表达与临床意义
Expression and clinical significance of miR-10b and HOXD10mRNA in human colorectal cancer

Abstract
Objective To investigate the expression of miR-10b and HOXD10mRNA in colorectal cancer tissues and to analyze their relation to clinical pathological characteristics.Methods Fifty three samples of colorectal cancer and pericancerous mucosal tissue were collected.The real-time PCR was used to detect the expression of miR-10b and HOXD10mRNA,and immunohistochemical SP method was used to detect the expression of HOXD10 protein in cancer and normal tissues.SPSS21.0 statistical software was applied for data analysis.Results In 53 colorectal cancer patients,miR-10b expression was elevated in 34 cases (65.38%);the relative expression of miR-10b was 1.672(1.167-1.945)and 0.648 (0.215-0.946) in colorectal cancer and normal mucosa tissues,respectively (P<0.05).The expression of HOXD10mRNA was reduced in 32 cases (60.38%);the relative expression of HOXD10mRNA was 0.341 (0.134-0.963) and 0.810(0.524-1.027) in colorectal cancer and normal mucosa tissues,respectively (P<0.05).The expression of HOXD10 protein was detected in 20 samples of cancer tissue (37.7%) and 41 samples of normal mucosa(77.4%)(P<0.05).The expression of miR-10b and HOXD10mRNA was significantly associated with lymph node metastasis and clinical stages (P<0.05),but not associated with age,sex,tumor sizes,local invasion.There was a negative correlation between miR-10b and HOXD10mRNA expression (P<0.05).Conclusion In colorectal cancer,the abnormal expression of miR-10b and HOXD10mRNA is related to invasion and metastasis of the cancer.The study indicates that miR-10b might be used as a novel target for biotherapy in colorectal cancer patients.关键词
