Effects of Variant Beclin Gene Expression on Cisplatin Sensitivity in Epithelial Ovarian Cancer Cells and Its Mechanism

摘要

Abstract

Objective To explore the effect of variant Beclin gene expression on cisplatin sensitivity of epithelial ovarian cancer cells and its mechanism. Methods The Beclin expression was detected in SKOV3 and SKOV3/DDP cells via RT-PCR. siRNA was used to in-hibit the Beclin expression in SKOV3 and SKOV3/DDP. The autophagy inhibitor 3-methyladenine (3-MA) was applied to interfere with the SKOV3/DDP cells. MTT method was applied to detect the sensitivity of cells to cisplatin. Results The expression of Beclin gene in SKOV3 resistant strains was significantly higher than in SKOV3 cells, and the difference was statistically significant (P<0.05). After siRNA inhibi-tion, the IC50of SKOV3, SKOV3/DDP, SKOV3si-Beclinand SKOV3/DDPsi-Beclinwere respectively (12.36±2.31)μg·mL-1, (17.31±1.91)μg·mL-1, (7.57±2.05)μg·mL-1and (12.46±1.58)μg·mL-1. The difference was statistically significant when compared with before siRNA inhibition (P<0.05).3-MA could inhibit the expression of Beclin in SKOV3/DDP cells and increase the sensitivity of cells to cisplatin. Conclusion The variant Beclin expression was directly associated with the sensitivity of SKOV3 cells to cisplatin, and this associationwas related to autophagy.
