IL-2对布鲁菌侵染小鼠巨噬细胞释放细胞因子及吞噬能力的调节作用
Effects of IL-2 on Release of Cytokines and Phagocytosis in Macrophages Infected with Brucella

Abstract
To investigate the regulatory effect of IL-2 on the release of cytokines (TNF-α,IFN-γ and NO) and phagocytic capacity of the macrophages infected with Brucella M5 strain,different doses of IL-2 were added to infected MΦ,and the content changes of TNF-α,IFN-γand NO released by MΦ(infection and no infection)infected with M5 at different time points were detected with ELISA kit;the number of intracel-lular viable bacteria and the phagocytic index of macrophages were calculated by CFU coating method in order to explore the effect of IL-2 on the immune regulation of MΦ after M5 infection.The test results showed that the cytokines (TNF-α,IFN-γ and NO)were released by M5 infected MΦ,and the levels of cytokines were significantly increased compared with the control group (P<0.05)under the regulation of IL-2,and there was a certain concentration and time dependence;IL-2 could promote the phagocytosis of macrophages to a certain extent,and the number of intracellular viable bacteria was also increased with the increase of IL-2 concentration,which was concentration-dependent.The results of the present study showed that IL-2 can promote the activation of MΦ and the secretion of TNF-α,IFN-γand NO by MΦ and enhance the immune response of the body.It may provide a new idea and theoretical basis for the clinical use of IL-2 and treatment of brucellosis.关键词
