维生素C对Treg/Th17失衡的调节作用
The effect of vitamin C on Treg/Th17 imbalance

Abstract
Objective:To study the effect of vitamin C(Vit C) on the imbalance of Treg/Th17 in EL4 cells.Methods:The proliferation rate of EL4 cells treated by different concentrations of Vit C or prednisone was detected by CCK-8.Transforming growth factor(TGF)-β1,interleukin(IL)-6 and IL-23 were used to induce CD4+T cells differentiation into Th17.The mRNA expressions of retinoic acid receptor-related orphan nuclear receptor gamma t(RORγt) and fork head box protein-3(Foxp3) were determined by qPCR.The levels of IL-17A and IL-10 in culture supernatant were detected by enzyme-linked immunosorbent assay(ELISA).Results:Both 0.2 mmol/L Vit C and 1 μmol/L prednisone had no obvious effect on the proliferation of EL4 cells,and could lead to increased expressions of RORγt and Foxp3,but a decreased secretion of IL-10(P<0.05).Conclusion:Vit C has a similar regulation effect on Treg/Th17 imbalance to that of prednisone(1 μmol/L),and there is a correlation between the intensity of the action and time.关键词
