Abstract

Aim:To investigate the effect of celastrol on proliferation,migration and apoptosis of human gastric cancer cell line HGC-27. Methods: MTT assay was used to detect the migration ability of HGC-27 cells treated with celastrol (0.0,0.5,1.0,1.5,2.0,3.0,4.0 μmol/L) for 24,48,72 h,respectively. The HGC-27 cells treated with celastrol(0.0, 0.5,1.0,1.5 μmol/L) for 48 h were to detect the migration ability by scratch test, apoptosis by flow cytometry, nuclear apoptotic bodies by Hochest staining, apoptosis and invasion related proteins Bcl-2, Bax, Cleaved Caspase-3, ERK, p-ERK, P38,and p-P38 by Western blot. Results:The proliferation ratio of HGC-27 treated with celastrol decreased signif-icantly,migration rate decreased,the early apoptotic rate increased(P<0.05) and nuclear apoptotic bodies formation was observed. The expressions of Bax,Cleaved Caspase-3 and p-P38 increased,while the expressions of Bcl-2 and p-ERK de-creased(P<0.05).Conclusion:Celastrol can inhibit the proliferation and migration of human gastric cancer cells and in-duce apoptosis.
