沉默单羧酸转运体4对宫颈癌Hela、Siha细胞增殖、侵袭迁移及凋亡的影响

Abstract
Aim:To investigate the effects of silencing monocarboxylate transporter 4(MCT4) gene on proliferation, invasion,migration and apoptosis of cervical cancer cells,Hela and Siha.Methods:The MCT4 gene of Hela and Siha cells were silenced by transient transfection of siRNA, and the siMCT4 sequence with good silencing effect was screened by Western blot for follow-up experiments. After the transfection,the lactic acid concentration in the extracellular fluid of each group was detected by the lactic acid kit. The cell proliferation of each group was detected by the CCK8 experiment. The cell migration was detected by the scratch test,and the invasion ability of the cells was detected by the Transwell invasion experiment. Apoptosis was detected by flow cytometry. Results:Western blot showed that the effect of siMCT4#2 was bet-ter in silencing MCT4 of Hela and Siha. Compared with the blank control group and the negative control group,after trans-fected with siMCT4#2 in Hela and Siha,the concentration of lactic acid in extracellular fluid,the cell migration rate, the invasion rate and cell proliferation rate all decreased in experimental group(siMCT4#2)(P<0.05),but the cell apopto-sis rate increased(P<0.001).Conclusion: Silencing MCT4 gene in Hela and Siha can inhibit the efflux of lactic acid, inhibit the proliferation,invasion and migration of two cell lines and promote their apoptosis.关键词
