Effect of Angiopoietin-like 4 on Vascular Permeability
摘要
Abstract
Objective To evaluate the effect of angiopoietin-like 4 (ANGPTL 4) on vascular permeability and the mechanisms.Methods Under normal oxygen or hypo-oxygen condition,human umbilical vein endothelial cells (HUVEC)were cultured in medium containing ANGPTL 4 at different concentrations and the permeability was detected as diffusion of FITC-BSA across HUVEC monolayer and distribution of [β-Catenin and ZO-1 by immunoflurescence assay.Results At any working concentration from 0.625 μtg/mL to 5 μg/mL,ANGPTL4 increased the HUVEC monolayer permeability,especially under hypo-oxygen condition (P＜0.05).ANGPTL4 promoted translocation of β-Catenin from cell surface to nucleus,and enlarged intercellular gap,which was accentuated under hypo-oxygen condition.Conclusions ANGPTL4 can increase HUVEC permeability effectively depending on concentration degree,especially under hypo-oxygen condition.This effect may be correlated to the interruption of intercellular connection.关键词
