海南橡胶树炭疽菌Colletotrichum siamense和C.fructicola的鉴定及系统发育分析
Identification and Phylogenetic Analysis of Anthracnose Pathogen Colletotrichum siamense and C.fructicola Isolated from Rubber Tree in Hainan

Abstract
Colletotrichum gloeosporioides species complex are reported as the main pathogen of the rubber anthracnose,which is one of the important leaf diseases of rubber tree in China.But accurate identification of the causal organisms is not known till now.In this study,thirteen strains of C.gloeosporioides species complex from the rubber tree in Hainan were selected for further species accurate identification.Partial mating type (Mat1-2) gene (ApMAT) and glutamine synthetase (GS) of them were cloned and the molecular phylogenetic tree of them were analyzed comparatively based on the partial sequences of ApMAT single gene,2-gene combined dataset (GS,ApMAT) and 6-gene combined dataset (ITS,ACT,CHS-1,GAPDH,GS,ApMAT),respectively.The results showed that the phylogenetic trees of the 13 isolates belong to two groups of C.siamense and C.fructicola,no matter the trees were builded using ApMAT single gene or 2-gene combined dataset (GS,ApMAT) or 6-gene combined dataset.It indicated that the 13 isolates of C.gloeosporioides species complex from the rubber tree in Hainan were identified as C.siamense and C.fructicola.In addition,our study demonstrated that the combined use of ApMat and GS in a phylogenetic analysis can identify the species in the C.gloeosporioides species complex.关键词
