西葫芦细菌性茎基软腐病病原的分离与鉴定
Isolation and identification of pectobacterium carotovorum subsp.carotovorum,the causal agent of stem soft rot on cucurbita pepo L.
摘要
Abstract
[Objective]Potential pathogens causing squash stem soft rot disease on greenhouse vegetables of Shanxi prov-ince were isolated and identified.[Methods]Forty isolated strains from squash were analyzed and identified by patho-genicity identification,morphological observation,determination of physiological and biochemical characteristics,and the analysis of 16SrDNA sequence.[Results]Pathogenicity test of strains isolated from squash revealed that CY 39 strain induced the disease agent.Further identification with physiological properties,16S rDNA sequence,and PCR-RFLP confirmed that isolates was P.carotovorum subsp.carotovorum(Pcc).The 16S rDNA testing of isolates shared 99% ~100% sequence similarity with that of Pcc strains.The phylogenetic trees showed that the isolates were clus-tered under the group of Pcc and P.chrysanthemi.and the sequence similarity between CY39 strain and AB680280 of Pectobacterium carotovorum subsp.Carotovorum was reached up to 100%.[Conclusion]To our best knowledge,this is the first report that Pcc has been proven as a causal agent of soft -rot disease on Cucurbita pepo L.关键词
