Abstract

[Objective]Present study was conducted to better understanding the MYB gene family and to explore the re-lationship between MYB gene family and sex differentiation of grape floral organs which would provide evidence for fur-ther research on sex differentiation of floral organs in grape.[Methods]Bisexual flowers of 'Beichun'grape species at full-blooming stage(corolla was off),single-male flowers of '520A'grape species,and female flowers of'1316C'grape species were selected as testing materials.Three repelications were set up.RNA was extracted from each sample to an-alyze transcriptome sequencing,and bioinformatics analysis.[Results]Four genes were expressed in the genomes of different sex flower organs,and the expression of the V IT_14s0066g01090 was the highest.V IT_13s0067g01630 and V IT_12s0059g00700 expression in single male flowers were significantly higher than that in bisexual flowers and '1316C'female flowers,while no significant difference of gene expression was detected between bisexual flowers and fe-male flowers.The expression level of V IT_19s0015g01280 was the highest in female flowers and significantly higher than that in bisexual flower,however there was no significant difference of the expression of V IT_19s0015g01280 be-tween female flowers and single male flowers.The phylogenetic tree showed that VIT_19s0015g01280 and AtMYB80 were clustered into one branch.[Conclusion]It was preliminarily determined that some genes from MYB gene family were involved in regulating sex differentiation of grape floral organs,which were the candidate genes to control sex dif-ferentiation of floral organs.The present study provided a theoretical reference for further research on sex differentia-tion of floral organs in grape.
